چکیده

Abstract Aquaporins are a family of ubiquitously expressed transmembrane water channels. We have previously reported that Aquaporin 4 (AQP4) is on T cells and treatment with small molecule AQP4 inhibitor significantly delays cell mediated heart allograft rejection. The goal this study was to identify mechanisms prolonged graft survival following blockade. Stimulation murine WT + AER-270 or −/−T resulted in decreased expression activation markers CD25 CD44, reduced volume when compared control cells. In addition, the absence inhibition also limited IL-2 production by stimulation. Phosphorylation of rat aquaporin-4 at Ser(111) is not required for channel gating.

Aquaporin 4 (AQP4) is the predominant water channel in the mammalian brain and is mainly expressed in the perivascular glial endfeet at the brain-blood interface. AQP4 has been described as an important entry and exit site for water during formation of brain edema and regulation of AQP4 is therefore of therapeutic interest. Phosphorylation of some aquaporins has been proposed to regulate their ...

Caspase Activation Is Required for T Cell Proliferation

Triggering of Fas (CD95) by its ligand (FasL) rapidly induces cell death via recruitment of the adaptor protein Fas-associated death domain (FADD), resulting in activation of a caspase cascade. It was thus surprising that T lymphocytes deficient in FADD were reported recently to be not only resistant to FasL-mediated apoptosis, but also defective in their proliferative capacity. This finding su...

Nuclear calcium is required for human T cell activation

Calcium signals in stimulated T cells are generally considered single entities that merely trigger immune responses, whereas costimulatory events specify the type of reaction. Here we show that the "T cell calcium signal" is a composite signal harboring two distinct components that antagonistically control genomic programs underlying the immune response. Using human T cells from healthy individ...

Antigen exposure required for T cell activation

Background A high frequency of hypersensitivity reactions to -lactam antibiotics are observed in patients with cystic fibrosis. -lactam antibiotics form protein conjugates in vitro and in vivo, and a core group of lysine residues of human serum albumin have been shown to be penicilloylated by mass spectrometric methods. Furthermore, protein/ peptide conjugates have been shown to stimulate T cel...

Rab11 is required for maintenance of cell shape via βPS integrin mediated cell adhesion in Drosophila

In eukaryotes, vesicle trafficking is regulated by the small monomeric GTPases of the Rab protein family. Rab11, (a subfamily of the Ypt/Rab gene family) an evolutionarily conserved, ubiquitously expressed subfamily of small monomeric Rab GTPases, has been implicated in regulating vesicular trafficking through the recycling of endosomal compartment. In an earlier communication, we have shown th...
